During ontogeny of Gossypium hirsutum L. floral buds (squares), increases in area and dry mass (DM) of floral bracts and the
subtending sympodial leaf followed a sigmoid growth curve with increasing square age. The maximum growth rates of the bract
area and bract DM occurred between 15 and 20 d after square first appearance (3 mm in diameter). Net photosynthetic rate (PN) of the sympodial leaf at first fruiting branch position of main-stem node 10 reached a maximum when the subtended square
developed into a white flower. Floral bracts had much lower PN and higher dark respiration than the subtending leaf. The amount of 14CO2 fixation by the bracts of a 20-d-old square was only 22 % of the subtending leaf, but 56 % of 14C-assimilate in the floral bud was accumulated from the bracts, 27 % from the subtending leaf, and only 17 % from the main-stem
leaf at 6 h after 14C feeding these source s. Hence floral bracts play an important role in the carbon supply of developing cotton squares.

High temperature limits in vivo pollen tube growth rates by altering diurnal carbohydrate balance in field-grown Gossypium hirsutum pistils 总被引:1,自引:0,他引:1
It has recently been reported that high temperature slows in vivo pollen tube growth rates in Gossypium hirsutum pistils under field conditions. Although numerous physical and biochemical pollen-pistil interactions are necessary for in vivo pollen tube growth to occur, studies investigating the influence of heat-induced changes in pistil biochemistry on in vivo pollen tube growth rates are lacking. We hypothesized that high temperature would alter diurnal pistil biochemistry and that pollen tube growth rates would be dependent upon the soluble carbohydrate content of the pistil during pollen tube growth. G. hirsutum seeds were sown on different dates to obtain flowers exposed to contrasting ambient temperatures but at the same developmental stage. Diurnal pistil measurements included carbohydrate balance, glutathione reductase (GR; EC 1.8.1.7), soluble protein, superoxide dismutase (SOD; EC 1.15.1.1), NADPH oxidase (NOX; EC 1.6.3.1), adenosine triphosphate (ATP), and water-soluble calcium. Soluble carbohydrate levels in cotton pistils were as much as 67.5% lower under high temperature conditions (34.6 °C maximum air temperature; August 4, 2009) than under cooler conditions (29.9 °C maximum air temperature; August 14, 2009). Regression analysis revealed that pollen tube growth rates were highly correlated with the soluble carbohydrate content of the pistil during pollen tube growth (r2 = 0.932). Higher ambient temperature conditions on August 4 increased GR activity in the pistil only during periods not associated with in vivo pollen tube growth; pistil protein content declined earlier in the day under high temperatures; SOD and NOX were unaffected by either sample date or time of day; pistil ATP and water soluble calcium were unaffected by the warmer temperatures. We conclude that moderate heat stress significantly alters diurnal carbohydrate balance in the pistil and suggest that pollen tube growth rate through the style may be limited by soluble carbohydrate supply in the pistil. 相似文献

Within SPIDIA, an EC FP7 project aimed to improve pre analytic procedures, the PAXgene Tissue System (PAXgene), was designed to improve tissue quality for parallel molecular and morphological analysis. Within the SPIDIA project promising results were found in both genomic and proteomic experiments with PAXgene-fixed and paraffin embedded tissue derived biomolecules. But, for this technology to be accepted for use in both clinical and basic research, it is essential that its adequacy for preserving morphology and antigenicity is validated relative to formalin fixation. It is our aim to assess the suitability of PAXgene tissue fixation for (immuno)histological methods. Normal human tissue specimens (n = 70) were collected and divided into equal parts for fixation either with formalin or PAXgene. Sections of the obtained paraffin-embedded tissue were cut and stained. Morphological aspects of PAXgene-fixed tissue were described and also scored relative to formalin-fixed tissue. Performance of PAXgene-fixed tissue in immunohistochemical and in situ hybridization assays was also assessed relative to the corresponding formalin-fixed tissues. Morphology of PAXgene-fixed paraffin embedded tissue was well preserved and deemed adequate for diagnostics in most cases. Some antigens in PAXgene-fixed and paraffin embedded sections were detectable without the need for antigen retrieval, while others were detected using standard, formalin fixation based, immunohistochemistry protocols. Comparable results were obtained with in situ hybridization and histochemical stains. Basically all assessed histological techniques were found to be applicable to PAXgene-fixed and paraffin embedded tissue. In general results obtained with PAXgene-fixed tissue are comparable to those of formalin-fixed tissue. A porous-membrane technique has been successfully used in root studies of field-grown soybean,Glycine max L. Merr., for several years. In order to evaluate this technique on other crops, a study was conducted to compare growth parameters of individual soybean, cotton,Gossypium hirsutum L., and corn,Zea mays L., plants grown with and without their root systems confined within porous membranes. Results indicate that plant species respond differently to root confinement.Root confinement reduced all shoot growth and yield parameters of the crop species, however the reduction was generally greatest for cotton. Apparently, cotton is less adaptable to the restricted rooting volume. Root confinement reduced levels of potassium and phosphorus in soybean and nitrogen in corn and cotton. Fertilizer rates higher than soil test recommendations should be applied to membrane-grown plants to prevent reductions in nutrient levels. It may be possible that modifying the size or shape of the membrane may enhance root and shoot growth of specific crop plants like cotton and increase the ability of this technique to be used on a broader spectrum of crop plants. 相似文献

The measurement of serum and urinary estrone sulfate (ES) was evaluated on random single samples for the detection of pregnancy in the Przewalski's horse (PRZ) and Hartmann's mountain zebra (HMZ). Pregnancy was detected by ES analysis of urine or serum in the PRZ and HMZ by at least 8 and 10 months before delivery, respectively. The values of ES in both serum and urine are shown at various stages of gestation. Results indicate urinary ES analysis is a noninvasive early diagnosis for pregnancy in these species. 相似文献

Using in vitro systems, numerous authors have cited the sensitivity of pollen tube growth to high temperature as a major cause of low yields for crops with valuable reproductive structures. We investigated the hypothesis that in vivo fertilization efficiency would be negatively affected by heat stress-induced changes in energy reserves and calcium-mediated oxidative status in the pistil. Gossypium hirsutum plants exposed to optimal (30/20°C) or high day temperature (38/20°C) conditions during flowering were analyzed for fertilization efficiency via UV microscopic observation of pollen tube-containing ovules and for soluble carbohydrates, adenosine triphosphate (ATP), calcium, antioxidant enzyme activity and NADPH oxidase (NOX; EC 1.6.3.1) activity in the pistil. Leaf measurements included gas exchange, chlorophyll content, quantum efficiency and ATP content of the subtending leaf on the day of anthesis. In the pistil fertilization efficiency, soluble carbohydrates, ATP content and NOX activity declined significantly, whereas water soluble calcium and glutathione reductase (EC 1.8.1.7) activity increased, and superoxide dismutase (EC 1.15.1.1) activity remained unchanged. In leaves, heat stress decreased photosynthesis, quantum efficiency and chlorophyll content, but increased stomatal conductance. We conclude that decreased source leaf activity either inhibits pollen development, tube growth through the style or guidance to the ovules as a result of an insufficient energy supply to the developing pistil. We further conclude that a calcium-augmented antioxidant response in heat-stressed pistils interferes with enzymatic superoxide production needed for normal pollen tube growth and fertilization of the ovule. 相似文献
